ENPP1 belongs to the ecto-nucleotide pyrophosphatase/phosphodiesterase (ENPP) family. ENPP1 is a type II transmembrane glycoprotein with two identical disulfide-bonded subunits. The protein's specificity is wide, cleaving a number of substrates including nucleotide phosphodiester bonds and nucleotide sugars as well as pyrophosphate links of nucleotides and nucleotide sugars. It can hydrolyze nucleoside 5' triphosphates to their monophosphates and diadenosine polyphosphates. ENPP1 has been linked to idiopathic infantile arterial calcification, ossification of the posterior longitudinal ligament of the spine (OPLL), and insulin resistance. ENPP1 is the most common cause of iduronidase deficiency and a variety of other diseases, including Cole disease and Hypophosphatemic Rickets.
