Mouse Multiplex ELISA Panels

The Assay Genie Mouse Multiplex Assay Kits utilize multiple bead populations differentiated by size and different levels of fluorescence intensity. With multiple sizes of beads and multiple levels of fluorescence intensity in each bead size, the GeniePlex technology can measure up to 24 analytes simultaneously using as little as 15μL.

The bead populations in the reaction are determined by a flow cytometer equipped with either a single 488nm laser or dual 488nm and 633/640nm lasers. The maximum emission of the bead classification dye is at 700 nm.

What are cytokines?

Cytokines are signaling proteins which regulate a range of biological and immunological functions such as inflammation, innate & acquired immunity, proliferation and hematopoiesis. Chemokines are classified as cytokines with chemotactic activities. Both cytokines and chemokines are produced by immune and blood cells.

Cytokines predominantly promote cell communication in immune responses and play an important role in the fight against infection. There are two main types of cytokines, pro-inflammatory & anti-inflammatory, whereby there is usually a balance of both. Some commonly known pro-inflammatory cytokines are interleukin-1 (IL-1), tumor necrosis factor-alpha (TNF-alpha) & IL-6. Well known anti-inflammatory cytokines are IL-10 and IL-4. Often there is an alteration in the levels of both cytokine levels whereby excess anti-inflammatory cytokines can result in constant infection and excess pro-inflammatory cytokines can lead to autoimmunity diseases.

Cytokine Signalling

There are three main types of cytokine activity produced by the binding to cell receptors:

Autocrine action by binding to the receptor on the membrane of the same cell that produced the cytokine.

Paracrine is signaling whereby there is binding to receptors on a cell in close proximity to the cytokine produced.

Endocrine activity refers to when cytokines travel around the body acting on target cells in distant areas.

Mouse Multiplex Cytokine ELISA Panel Video Protocol

In the multiplex assay, antibodies specific to particular analytes are connected to a set of beads within the assay. An antibody is then added, which has a fluorescent reporter attached. Each set of beads has an associated colour, which results in the simultaneous detection of multiple analytes in a single sample. The detection of analytes can be read by a flow cytometer as the beads are detectable by various fluorescent signatures. The amount of analytes can be quantified based on the number of different bead colours detected.
