Ankyrin-R (ANK1), originally identified in erythrocytes, is a member of the ankyrin family of adaptor proteins that tether integral membrane proteins to the spectrin-actin cytoskeleton. This structural linkage is essential for maintaining plasma membrane stability and for the precise localization of ion channels, exchangers, and transporters across various cell types. Although Ankyrin-R is best known for its role in red blood cells—where mutations cause hereditary spherocytosis—it is also expressed in the brain and muscle tissue. In neurons, Ankyrin-R contributes to the organization of membrane domains and supports the structural integrity of axons and dendrites. Its ability to bind β-spectrin and multiple membrane proteins suggests a broader role in maintaining neuronal polarity and excitability.
