Aquaporin 3 (AQP3) is a member of the aquaporin family of integral membrane proteins that facilitate the selective transport of water and small solutes across cell membranes. Unlike classical water channels, AQP3 also conducts glycerol and other small neutral molecules, classifying it as an aquaglyceroporin. It is primarily localized to the basolateral membrane of principal cells in the renal collecting duct, where it enables water efflux following vasopressin-induced apical water reabsorption. Notably, AQP3 expression is not directly regulated by vasopressin. Beyond its established role in renal physiology, AQP3 is increasingly recognized for its relevance in the central nervous system. It is expressed in brain endothelial cells, astrocytes, and certain neuronal populations, where it contributes to water and glycerol homeostasis, oxidative stress regulation, and cell volume control. These functions are critical in maintaining blood-brain barrier integrity and responding to neuroinflammatory and ischemic insults. Emerging evidence suggests that dysregulation of AQP3 may be involved in the pathogenesis of neurodegenerative diseases such as Alzheimer’s disease and Parkinson’s disease, where altered fluid dynamics, oxidative stress, and metabolic dysfunction are key pathological features. AQP3’s dual role in water and glycerol transport positions it as a potential modulator of neuronal resilience and metabolic adaptation.
