The activity of mitochondrial Complex I can be quantified by measuring the change in optical density at 340 nm, reflecting NADH consumption during the enzymatic reaction.
This kit employs a detection principle whereby mitochondrial Complex I catalyzes the oxidation of NADH to NAD+. The activity of mitochondrial Complex I is determined by measuring the change in optical density (ΔOD) at 340 nm over a 5-minute incubation period at 37 °C, which reflects the consumption of NADH substrate by the enzyme.
