ClC-3 (ClCN3) is a voltage-gated chloride/proton exchanger that plays a critical role in regulating endosomal and synaptic vesicle acidification. By mediating Cl⁻/H⁺ exchange, ClC-3 contributes to vesicle trafficking, neurotransmitter release, and membrane excitability—key processes in neuronal communication and plasticity. In the brain, ClC-3 is expressed in neurons and glial cells, where it influences synaptic vesicle cycling and endolysosomal function. Dysregulation of ClC-3 has been linked to impaired short-term memory, altered neuronal excitability, and neurodegenerative phenotypes in animal models. Its modulation by protein kinase C suggests a role in activity-dependent signaling pathways. Given its involvement in vesicle dynamics and ion homeostasis, ClC-3 is emerging as a potential target in neurodegenerative disease research, particularly in disorders characterized by synaptic dysfunction and lysosomal impairment.
