NR2B (GRIN2B) is a subunit of the NMDA receptor complex, predominantly expressed during early brain development and in specific regions of the adult brain such as the hippocampus and cortex. NR2B-containing NMDA receptors exhibit slower deactivation kinetics and higher calcium permeability, making them critical for synaptic plasticity, memory formation, and neurodevelopment. In neurodegenerative disease research, NR2B has been implicated in excitotoxicity, a pathological process driven by excessive glutamate signaling and calcium influx. Overactivation of NR2B-containing receptors contributes to neuronal injury in conditions such as Alzheimer’s disease, Parkinson’s disease, and stroke. Pharmacological modulation of NR2B has shown promise in preclinical models for reducing neurotoxicity and improving cognitive function. NR2B-selective antagonists are being explored as therapeutic agents for neuroprotection and symptom management in neurodegenerative and neuropsychiatric disorders.
