Human NEFL (Neurofilament, Light Polypeptide) ELISA Kit
The Human NEFL (Neurofilament, Light Polypeptide) ELISA Kit is designed for the quantitative detection of NEFL levels in human serum, plasma, cell supernatant, lysates, and other biological samples. Neurofilament Light Chain (NEFL or NfL) is a 68 kDa cytoskeletal intermediate filament protein expressed predominantly in neurons, where it forms heteropolymers with neurofilament medium (NEFM) and heavy (NEFH) subunits to maintain axonal caliber, structural integrity, and conduction velocity. Upon axonal injury or neuronal degeneration, NEFL is released into the cerebrospinal fluid and bloodstream, making it a sensitive and specific blood-based biomarker of neuroaxonal damage. Elevated NEFL levels have been documented across a wide spectrum of neurological disorders, including multiple sclerosis, Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis (ALS), frontotemporal dementia, traumatic brain injury, stroke, and Charcot-Marie-Tooth disease. Accurate measurement of NEFL is essential for understanding neurodegenerative disease mechanisms and for developing targeted therapeutic strategies. Assay Genie's Human NEFL ELISA Kit offers exceptional sensitivity and specificity, ensuring reliable and reproducible results. Manufactured under stringent quality control standards, this kit provides robust performance and is easy to use, making it an excellent choice for both research and translational applications. Trust Assay Genie's Human NEFL ELISA Kit for accurate and dependable quantification of this crucial biomarker in your studies.

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human NEFL. Standards or samples are added to the wells, allowing the target to bind to the immobilized antibody. After incubation, a biotinylated detection antibody specific for Human NEFL is added, which binds specifically to the captured target. Following a wash step to remove excess detection antibody, HRP-conjugated Streptavidin is introduced, forming a biotin-streptavidin-HRP complex. After a second washing step, Substrate Reagent is added to initiate a colorimetric reaction catalyzed by HRP. The reaction produces a blue product that turns yellow upon addition of the acidic Stop Solution. The optical density (OD) is measured at 450 nm using a microplate reader. The OD450 value is directly proportional to the concentration of NEFL in the sample, which can be determined by referencing a standard curve.
