JC-10 is a dual-emission ratiometric probe that measures mitochondrial membrane potential. In healthy cells with high mitochondrial membrane potential, JC-10 concentrates inside mitochondria and forms J-aggregates, emitting orange fluorescence (540 nm / 590 nm). In stressed or depolarised cells where membrane potential collapses, JC-10 disperses into the cytoplasm as monomers, shifting fluorescence to green (490 nm / 525 nm). The ratio of orange-to-green fluorescence (F590 / F525) directly measures mitochondrial depolarisation.

| Positive Control (FCCP): | Add 10 µl of 40 µM FCCP to designated wells; incubate 30 min at 37°C |
