Smith (Sm) Antigen (SmA) Human IgA ELISA Kit (SBRS8818)
The Smith (Sm) Antigen (SmA) Human IgA ELISA Kit is an indirect, colorimetric enzyme-linked immunosorbent assay for the detection of human IgA antibodies against Smith (Sm) antigen in serum and plasma samples. Plates are pre-coated with Sm antigen, and a Positive Control containing anti-antigen antibody is supplied to generate a calibration curve for result interpretation.
Antigen-specific IgA in the sample binds to the coated wells and is detected with a biotinylated anti-human IgA antibody, followed by HRP-Streptavidin and TMB substrate. The Stop Solution changes the color from blue to yellow, and the intensity, read at 450 nm, is proportional to the amount of anti-Sm antigen IgA present.

- Prepare all reagents, samples and positive control calibration curve as instructed.
- Add 100 µl positive control or sample (diluted 1:1500) to each well.
- Incubate 1 hour at room temperature with gentle shaking.
- Add 100 µl prepared Biotinylated Anti-Positive Control and Biotinylated Anti-Human IgA Antibody into respective wells.
- Incubate 1 hour at room temperature with gentle shaking.
- Add 100 µl prepared HRP-Streptavidin solution to each well.
- Incubate 30 minutes at room temperature with gentle shaking.
- Add 100 µl TMB One-Step Substrate Reagent to each well.
- Incubate 30 minutes at room temperature in the dark with gentle shaking.
- Add 50 µl Stop Solution to each well.
- Read at 450 nm immediately.
The entire kit may be stored at -20°C for up to 1 year from the date of shipment. Avoid repeated freeze-thaw cycles. The kit may be stored at 4°C for up to 6 months. For extended storage, it is recommended to store at -80°C. For prepared reagent storage, see the technical manual.
