Thyroid hormone receptors (TRs) are ligand-dependent transcription factors belonging to the nuclear receptor superfamily, which also includes steroid and retinoic acid receptors. Activated by triiodothyronine (T3), TRs regulate the expression of genes involved in metabolism, growth, and cellular differentiation. In the central nervous system, TRs are essential for brain development, synaptic plasticity, and neurogenesis. During early neurodevelopment, thyroid hormones influence neuronal migration, myelination, and axonal growth. In the adult brain, TRs continue to modulate neurophysiological processes, including mitochondrial function, oxidative stress response, and neurotransmitter signaling.
