Description
ALP Detection Reagent (BA0169) (BA0169)
The ALP Detection Reagent (SKU: BA0169) is a stable, ready-to-use 4-methylumbelliferyl phosphate (MUP) liquid substrate reagent formulated for the detection of alkaline phosphatase activity, for example in ELISA-based assays. When dephosphorylated by alkaline phosphatase, this single reagent produces a highly fluorescent product that is measured at excitation/emission wavelengths of 360/450 nm. The fluorescent signal generated may also be visualised directly under a UV light source. Supplied as a 10x concentrated reagent, a single kit provides sufficient material for up to 600 assays in a 96-well plate format. The reagent is convenient to use, requiring only a simple ten-fold dilution in distilled water prior to application. It is intended for research use only.

A stable 4-methylumbelliferyl phosphate liquid substrate reagent for the fluorimetric detection of alkaline phosphatase activity. Upon dephosphorylation by ALP, the reagent yields the highly fluorescent product 4-methylumbelliferone, which is measured at 360/450 nm and may also be visualised under UV light. The reagent is supplied as a 10x concentrate and is sufficient for 600 assays in 96-well plate format.

Note: The below protocol is a sample protocol. Protocols are specific to each batch/lot. For the correct instructions please follow the protocol included in your kit.
| Step | Procedure |
| 1 | Prior to use, dilute the reagent ten-fold in distilled water. |
| 2 | For ELISA, add 100 uL of diluted reagent per well in a 96-well plate. |
| 3 | Incubate for 30 minutes at room temperature. |
| 4 | If desired, stop the reaction by adding 100 uL phosphate-buffered saline (1x PBS). |
| 5 | Read fluorescence at 360/450 nm. |
| Component | Quantity | Storage |
