DNA and RNA damage are critical molecular events that contribute to the onset and progression of neurodegenerative diseases. These lesions arise from both endogenous metabolic processes and environmental stressors, including oxidative stress, inflammation, and exposure to neurotoxins. In neurons—cells with limited regenerative capacity—such damage can have profound and lasting effects. Reactive oxygen and nitrogen species (RONS), such as hydroxyl radicals and peroxynitrite, induce oxidative modifications to nucleic acids. Key biomarkers of this damage include 8-hydroxyguanine (8-OHG), 8-hydroxy-2'-deoxyguanosine (8-OHdG), and 8-hydroxyguanosine (8-OHG), which are associated with mutagenic base transversions and impaired transcriptional fidelity. These lesions disrupt gene expression, mitochondrial function, and synaptic signaling—hallmarks of neurodegenerative pathology.
