DNMT1 (DNA methyltransferase 1) is the primary enzyme responsible for maintaining DNA methylation patterns during cell division. In the nervous system, DNMT1 regulates gene expression, synaptic plasticity, and neuronal differentiation through epigenetic mechanisms. Aberrant DNMT1 activity has been linked to neurodegenerative diseases, where altered methylation contributes to transcriptional silencing of neuroprotective genes and activation of pathogenic pathways. DNMT1 interacts with key regulatory proteins such as retinoblastoma (Rb), E2F1, and HDAC1, forming complexes that influence chromatin structure and gene repression. Loss of DNMT1 function leads to global hypomethylation, genomic instability, and impaired neuronal survival. Its role in epigenetic remodeling makes DNMT1 a promising target for therapeutic intervention and biomarker discovery in neurodegeneration.
