Description
dsDNA high-sensitivity assay kit
GenieQuant 1× dsDNA HS Assay Kit is a fluorometric assay for highly sensitive, selective quantification of double-stranded DNA, accurate from picogram levels across a low, narrow concentration range. It is ideal for NGS library QC, cfDNA and low-input samples. The working solution is supplied ready to use (1×), so you simply add sample.
The kit contains a premixed working solution with a dsDNA-specific fluorescent dye, plus two dsDNA standards. Fluorescence is linear from 0.2 to 100 ng dsDNA, allowing accurate quantification of sample concentrations from 10 pg/μl to 100 ng/μl. The dye reaches saturation within 2 minutes and tolerates common contaminants such as RNA, salts, free nucleotides, proteins, solvents and detergents. The protocol is designed for Qubit 2.0, 3.0 and 4.0 fluorometers. For research use only.

Storage
Store at 2 ~ 8°C.
This protocol is for Qubit 2.0, 3.0 and 4.0 fluorometers only.
- Equilibrate
- Bring all components to room temperature before use.
- Prepare tubes
- Use 0.5 ml PCR tubes only, one for each sample and standard.
- Label the lids, not the sides, as side labels can interfere with the reading.
- Prepare the standards
- Add 190 μl of working solution to each standard tube, then add 10 μl of Standard #1 or Standard #2 to the corresponding tube.
- Vortex gently for 2–3 sec, avoiding bubbles. Pipette accurately.
- Prepare the samples
- Add 180–199 μl of working solution to each sample tube, then add 1–20 μl of dsDNA sample to a final volume of 200 μl.
- Vortex gently for 2–3 sec, avoiding bubbles. Pipette accurately.
- Incubate
- Incubate all tubes at room temperature for 2 min, protected from light.
- Read
- Select the dsDNA High Sensitivity assay program on the fluorometer and measure the concentration according to the instrument instructions.

Q1: What concentration range can the kit measure?
- The assay is linear from 0.2 to 100 ng dsDNA in the tube, corresponding to sample concentrations of 10 pg/μl to 100 ng/μl.
Q2: Will RNA or other contaminants affect the result?
- The dye is specific for dsDNA and tolerates common contaminants such as RNA, salts, free nucleotides, proteins, solvents and detergents.
Q3: Which instruments and tubes are compatible?
- The protocol is designed for Qubit 2.0, 3.0 and 4.0 fluorometers, using 0.5 ml PCR tubes only.
Q4: How long are prepared tubes stable?
- Read samples within 3 h of adding them to the working solution to avoid fluorescence quenching.
