Description
Dilution buffer for NGS libraries
GenieQuant Library Dilution Buffer is a ready-to-use buffer for preparing serial dilutions of NGS libraries and standards ahead of qPCR-based library quantification. It is formulated to stabilise dilute DNA for accurate, reproducible results.
When DNA is diluted in ddH2O or other buffers, especially to very low concentrations, it can adsorb to tubes and similar containers, making dilutions inaccurate. The optimised components of this buffer prevent DNA from binding to tube walls, enabling accurate dilution over a wider range. It is designed for diluting next-generation sequencing (NGS) libraries for library quantification and for preparing standard curves. For research use only.

Storage
Store at -30 ~ -15°C.
- Thaw and mix
- Thaw the buffer completely and mix thoroughly. It can then be used directly to dilute DNA libraries or templates.
- Perform serial dilutions
- Carefully prepare a serial dilution based on the concentration of the library or template.
- Avoid large single-step dilutions (e.g. 10,000-fold); use several smaller dilutions instead. For a 10,000-fold dilution, perform two consecutive 100-fold dilutions.
- Use promptly
- Keep diluted samples on ice and mix thoroughly before use. Prepare dilutions fresh and do not store them for extended periods.
Q1: Why not dilute libraries in water?
- At very low concentrations, DNA diluted in ddH2O or other buffers can adsorb to tubes and containers, making dilutions inaccurate. This buffer prevents that binding, giving accurate dilutions over a wider range.
Q2: How should I make a 10,000-fold dilution?
- Avoid a single large-volume step. Perform two consecutive 100-fold dilutions instead, because concentrated DNA is viscous and disperses poorly.
Q3: Can I store diluted libraries?
- No. Prepare dilutions fresh, keep them on ice and mix thoroughly before use; do not store them for extended periods.
