Description

Human HMBS ELISA Kit

Human HMBS ELISA kits are used to detect Hydroxymethylbilane Synthase (HMBS) in tissue homogenates, cell lysates and other biological samples. HMBS, also known as porphobilinogen deaminase, is the third enzyme of the haem biosynthetic pathway and is expressed in a housekeeping form and an erythroid-specific form. It catalyses the sequential head-to-tail polymerisation of four molecules of porphobilinogen to form the linear tetrapyrrole hydroxymethylbilane, also called preuroporphyrinogen. Catalysis begins with assembly of a dipyrromethane cofactor, which stays covalently bound and acts as a primer around which the tetrapyrrole is built, and the product is released at the end. Heterozygous loss-of-function variants cause acute intermittent porphyria, an autosomal dominant hepatic porphyria with attacks of abdominal pain, neurological dysfunction, hypertension and tachycardia, in which porphyrin precursors accumulate and are excreted in urine. HMBS activity and gene variants are therefore studied in porphyria diagnosis.
