MDM2 (Mouse Double Minute 2 homolog) is an E3 ubiquitin ligase that negatively regulates the tumor suppressor p53, controlling its stability and transcriptional activity. In the nervous system, MDM2-p53 signaling is crucial for balancing neuronal survival and apoptosis. Overactivation of p53, due to MDM2 dysfunction, can lead to excessive neuronal loss—a feature observed in neurodegenerative diseases such as Parkinson’s and ALS. MDM2 also influences synaptic plasticity and neurogenesis, suggesting broader roles in cognitive function and brain repair. Targeting the MDM2-p53 axis offers a potential neuroprotective strategy by modulating stress responses and preventing p53-mediated neurotoxicity in degenerative conditions.
