Protein arginylation, specifically at the N-terminus, is a critical post-translational modification mediated by the enzyme ATE1. This process regulates protein turnover, stress response, and cytoskeletal organization. In neurons, N-terminal arginylation facilitates the clearance of misfolded proteins and modulates actin dynamics, both essential for synaptic integrity and plasticity. Emerging evidence suggests that dysregulation of arginylation contributes to neurodegenerative pathologies by impairing proteostasis and enhancing susceptibility to cellular stress. Arginylation also influences the unfolded protein response and autophagy, linking it to mechanisms underlying Alzheimer’s and Huntington’s disease.
