PSD95 (Postsynaptic Density Protein 95), also known as SAP90, is a scaffolding protein in the membrane-associated guanylate kinase (MAGUK) family. It organizes the postsynaptic density by anchoring receptors, ion channels, and signaling molecules at excitatory synapses. PSD95 contains three PDZ domains, an SH3 domain, and a guanylate kinase-like domain. The PDZ domains mediate interactions with NMDA receptors, potassium channels, and neuroligins, facilitating synaptic signaling and plasticity. PSD95 also binds neuronal nitric oxide synthase, linking it to redox signaling. Through its role in AMPA receptor trafficking via Stargazin and TARPs, PSD95 influences synaptic strength and experience-dependent plasticity. Mutations in PSD95 are associated with autism spectrum disorders and cognitive deficits, underscoring its importance in learning and memory and its relevance in neurodegenerative disease research.
