SSI family members are cytokine-inducible negative regulators of cytokine signaling. The expression of this gene is induced by various cytokines, including IL6, IL10, and interferon (IFN)-gamma. The protein encoded by this gene can bind to JAK2 kinase, and inhibit the activity of JAK2 kinase. Studies of the mouse counterpart of this gene suggested the roles of this gene in the negative regulation of fetal liver hematopoiesis, and placental development.

Western blot analysis of lysates from RAW 264.7 cells using SOCS3 Rabbit mAb (CAB21981) at1:2000 dilution incubated overnight at 4℃. Raw 264. 7 cells were treated with LPS (1 μg/ml) at 37℃ for 8 hours. Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution. Lysates/proteins: 25 μg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Enhanced Kit (AbGn00021). Exposure time: 180 s.
