Description
Adenylate Kinase (ADK) Activity Assay Kit – Luminescent
The Assay Genie Adenylate Kinase (ADK) Activity Assay Kit is a fast, luminescent assay for measuring adenylate kinase (myokinase, EC 2.7.4.3) activity in cell and tissue lysates, diluted serum and cell culture medium. ADK catalyses the reversible reaction 2 ADP ↔ ATP + AMP and plays a central role in cellular energy homeostasis and adenine nucleotide signalling.
In this kit, ADK in the sample converts ADP substrate into ATP, which is quantified with a luciferase/luciferin detection reagent and read as relative light units (RLU). The enzymatic reaction takes just 1 minute for lysates (5 minutes for medium and serum), making the kit well suited to energy metabolism studies and to monitoring ADK released into culture medium as a marker of cell membrane damage.

- Ultra-fast enzymatic step: 1 min for cell and tissue lysates
- Highly responsive luciferase-based ATP readout
- Works with lysates, diluted serum and cell culture medium
- Includes TCA Stop for precise, reproducible reaction timing
- Includes 10x Cell Lysis Solution for sample preparation
- ADK in the sample converts the supplied ADP substrate into ATP (2 ADP → ATP + AMP).
- The reaction is stopped with TCA Stop, the samples are clarified and diluted, and ATP is quantified with the ATP Assay Solution (luciferase/luciferin).
- Light output is read as RLU on a luminometer; ADK activity is expressed as ΔRLU (sample − control).
- Cells: ~105 freeze-thawed cells in ~100 µl ice-cold 1x Cell Lysis Solution, 5 min on ice.
- Tissue: ~2 mg tissue homogenised in 0.5 ml ice-cold 1x Cell Lysis Solution.
- Centrifuge at ~14,000 rpm for 5 min at 4 °C; adjust protein to 0.05–0.5 mg/ml (BCA).
- Serum: dilute 3-fold with ice-cold dH2O. Do not use plasma or undiluted serum.

What samples can I use with the ADK Activity Assay Kit?
Cell and tissue lysates, serum diluted 3-fold, and cell culture medium. Plasma and undiluted serum are not recommended.
How long does the adenylate kinase assay take?
The enzymatic reaction runs for 1 min (lysates) or 5 min (medium and serum) at 37 °C, followed by a short stop, centrifugation and luminescence read.
What equipment do I need?
A luminometer or luminescence plate reader, a 37 °C water bath, a refrigerated microcentrifuge and a clear 96-well plate.
