The Cytotoxicity Control Reagent (Saponin) (SKU: BA0005) is a 50 mg preparation of saponin supplied as a positive cytotoxic control for use with cell viability and cytotoxicity assays. Saponin is a detergent that permeabilises cell membranes and reliably induces cell death, making it an ideal reference for total lysis or maximal cytotoxicity controls. The reagent is conveniently reconstituted in phosphate buffered saline to give a 1% saponin stock and is typically used at a final concentration around 0.1%. It is designed to accompany colorimetric and fluorescent cell viability kits, where a dose-dependent cytotoxic response can be demonstrated. Saponin is a cytotoxic detergent used as a positive control reagent in cell viability and cytotoxicity assays. It permeabilises the cell membrane to induce cell death, providing a total-lysis or maximal-cytotoxicity reference point. The reagent can be conveniently reconstituted with 5 mL phosphate buffered saline to give a 1% saponin solution and is commonly used at a final concentration of approximately 0.1% saponin in the assay well. It has been characterised as a cytotoxic control with an IC50 of approximately 0.002 wt%.
