Accumulation of misfolded proteins triggers the unfolded protein response via three ER sensors - PERK, IRE1alpha and ATF6. PERK phosphorylates eIF2alpha to attenuate translation and induce ATF4 and CHOP, while IRE1alpha-XBP1 expands ER capacity; BiP/GRP78 is the master chaperone.
