Macroglobulinemia and neuropathy, hereditary sensory and autonomic, type V, are examples of diseases associated with STOML2 (Stomatin-like protein 2). Pathways related to STOML2 include neuroscience. STOML2 is also involved in signalling receptor binding and cardiolipin binding. STOML3 is an important paralog of STOML2. STOML2 is a mitochondrial protein that appears to control the formation and operation of mitochondria. STOML2 activates cardiolipin synthesis and localization by binding to cardiolipin-enriched membranes, recruiting and stabilizing prohibitin and other proteins. Because it may act in the organization of functional microdomains in mitochondrial membranes, STOML2 might play a role in mitochondrial function. STOML2 may be involved in various biological processes such as cell migration, cell proliferation, T-cell activation, calcium homeostasis, and stress response through its role in mitochondrial regulation. STOML2 is necessary for mitochondrial hyperfusion, a pro-survival cellular response to stress that results in greater ATP production by mitochondria. STOML2 may be involved in calcium homeostasis through suppression of mitochondrial calcium efflux. STOML2 may also affect the formation of functional domains at the plasma membrane, and it might participate in T-cell activation through binding to the T-cell receptor signalling complex and its regulation.
