The tumour suppressor p53 is stabilised and activated by stress-induced phosphorylation (e.g. Ser15), escaping MDM2-mediated degradation to transactivate targets such as p21, PUMA, NOXA and GADD45 that enforce cell-cycle arrest, senescence or apoptosis. p53 is the most commonly mutated gene in human cancer.
