The cGAS-STING axis senses cytosolic DNA: cGAS synthesises cGAMP to activate STING, driving TBK1-mediated phosphorylation of IRF3 (and NF-kB) and type I interferon induction. The pathway is a key node in innate immunity, autoinflammation and anti-tumour immunity.
