DUX4 (Double Homeobox 4) is a transcription factor implicated in gene regulation during early development. In adult tissues, aberrant expression of DUX4 is associated with toxic gain-of-function effects, particularly in facioscapulohumeral muscular dystrophy (FSHD). Recent studies suggest that DUX4 may also contribute to neurodegenerative processes by activating stress response genes, disrupting RNA metabolism, and inducing apoptosis. Its expression in differentiated tissues, including skeletal muscle and potentially the nervous system, highlights its pathological relevance beyond muscular dystrophy. DUX4’s ability to modulate transcriptional networks and trigger cell death pathways positions it as a candidate for further investigation in neurodegeneration, especially in disorders involving epigenetic dysregulation and inflammatory signaling.
