The Malachite Green Phosphate Assay Kit (SKU: BA0048) offers a simple, direct and automation-ready method for the quantitative colorimetric determination of inorganic phosphate directly in samples without any pretreatment. Phosphate is one of the most important ionic species in nature, present in all biological systems and a major constituent of minerals, fertilisers and industrial wastewater. Accurate phosphate determination therefore finds numerous applications in pharmacology, biomedical research, clinical chemistry, industrial process monitoring and environmental monitoring. The improved Malachite Green method utilises malachite green dye and molybdate, which form a stable coloured complex specifically with inorganic phosphate. The intensity of the colour, measured at 620nm, is directly proportional to the phosphate concentration. The optimised formulation substantially reduces interference by substances in raw samples and can be run in cuvette or 96-well plate format.

This kit measures phosphate ion directly in samples without pretreatment. The improved Malachite Green method utilises malachite green dye and molybdate, which form a stable coloured complex specifically with inorganic phosphate. The intensity of the colour, measured at 620nm, is directly proportional to the phosphate concentration in the sample.

| 1 | Bring reagents to room temperature and shake before use. |
| 2 | Set up standards and samples: transfer 50 µL Blank Control, Standard and samples separately in duplicate wells of a clear bottom 96-well plate. |
| 3 | Add 100 µL Reagent and tap lightly to mix. |
| 4 | Incubate 30 minutes at room temperature and read optical density at 620nm (600-660nm). |
Phosphate concentration = [(ODSAMPLE − ODBLANK) / (ODSTANDARD − ODBLANK)] × 0.28 (mg/dL), where ODBLANK, ODSTANDARD and ODSAMPLE are OD620nm values of Blank, Standard and Sample. Conversion: 1 mg/dL Pi equals 105.3 µM, 0.001% or 10 ppm.
