Voltage-dependent anion-selective channel protein 1 (VDAC1), also known as mitochondrial porin 1, is a pivotal protein embedded in the outer mitochondrial membrane. It serves as the primary gateway for the exchange of ions and metabolites between mitochondria and the cytosol, playing a critical role in cellular energy metabolism, calcium homeostasis, and apoptosis. VDAC1 also functions as a docking site for key regulatory proteins, including hexokinase and BCL2L1, linking mitochondrial activity to glycolysis and cell survival pathways. In the context of neurodegenerative diseases, VDAC1 has emerged as a key player in mitochondrial dysfunction—a hallmark of disorders such as Alzheimer’s, Parkinson’s, and Huntington’s disease.
