The extrinsic pathway is initiated when death ligands such as FasL and TRAIL engage their receptors, nucleating the death-inducing signalling complex (DISC) through FADD. DISC assembly drives caspase-8/-10 activation, which can directly cleave executioner caspases or amplify killing via BID-mediated crosstalk to the mitochondrial pathway, with cFLIP acting as a key checkpoint.
