Interleukin-1 beta (IL-1β) is a key pro-inflammatory cytokine encoded by the IL1B gene and produced primarily by activated macrophages, monocytes, and dendritic cells. It is initially synthesized as an inactive 31 kDa precursor (pro-IL-1β) that is cleaved by Caspase-1, following inflammasome activation, into its biologically active 17 kDa mature form. IL-1β signals through the IL-1 receptor (IL-1R) to activate downstream pathways including NF-κB and MAPK, leading to the induction of inflammation, fever, leukocyte recruitment, and the expression of additional pro-inflammatory mediators.
