SHANK3 is a multidomain scaffolding protein located at the postsynaptic density of excitatory synapses. It anchors glutamate receptors, ion channels, and signaling proteins to the actin cytoskeleton, playing a vital role in synaptic structure and function. SHANK3 is involved in receptor tyrosine kinase signaling, including the ERK-MAPK and PI3K pathways, which are essential for synaptic plasticity and neuronal survival. Mutations or deletions in SHANK3 are strongly associated with autism spectrum disorders and Phelan-McDermid syndrome, often resulting in impaired language and social communication. In neurodegenerative disease, SHANK3 dysfunction contributes to synaptic disassembly and cognitive decline. Its role in maintaining synaptic integrity and regulating intracellular signaling makes SHANK3 a critical target for understanding and treating synaptic pathologies in both developmental and degenerative brain disorders.
