Receptor tyrosine kinases transduce extracellular growth factor signals across the plasma membrane to drive proliferation, survival, motility, and metabolism, and their deregulation is a pervasive feature of human cancer. Ligand binding induces receptor dimerization and trans-autophosphorylation of cytoplasmic tyrosine residues, creating docking sites that nucleate downstream signaling through the RAS-MAPK and PI3K-AKT cascades. Within the ERBB/HER family, EGFR is activated by EGF-family ligands, and its autophosphorylation, detected as Phospho-EGFR, marks active signaling; HER2/ErbB2 has no known ligand and acts as the preferred heterodimerization partner that potentiates signaling, while the kinase-impaired HER3/ErbB3 couples strongly to PI3K and partners with HER2 to form a potent oncogenic unit. Beyond the HER family, the hepatocyte growth factor receptor c-Met drives invasive growth and scattering, the insulin-like growth factor receptor IGF-1R supports proliferation and survival and contributes to therapy resistance, the fibroblast growth factor receptor FGFR2 regulates growth and differentiation, and the vascular endothelial growth factor receptor VEGFR2 mediates angiogenic signaling that supports the tumor microenvironment. In cancer, these receptors are activated by overexpression, gene amplification, activating mutation, and autocrine ligand loops: EGFR mutations and HER2 amplification define major therapeutic subsets in lung and breast cancer, while c-Met, FGFR2, and IGF-1R contribute to oncogenesis and to resistance against other targeted agents. This biology underpins a large class of small-molecule kinase inhibitors and therapeutic antibodies.
