Interferon-induced 17 kDa protein (ISG15), a 15-kDa protein with a unique primary amino acid sequence, is an intracellular ubiquitin homologue and cytokine that promotes IFN-gamma production and NK / lymphokine-activated killer cell proliferation and function. ISG15 is produced by monocytes and lymphocytes. ISG15 is a ubiquitin-like protein that is strongly upregulated as a primary response to various microbial and cellular stressors. Alterations in the ISG15 signalling pathway have also been found in several human tumour entities. ISG15 expression is also significantly increased by viral or bacterial infection via the Janus kinase/signal transducer and activator of transcription (Jak / STAT) signalling pathway, in addition to being induced by type I interferon. ISG15 is secreted by monocytes, B- and T-lymphocytes, and fibroblasts after induction. We show that the purpose of the ISG15 protein is blocked by influenza B virus in a novel way, which strongly activates the ISG15 protein: an area of the influenza B virus NS1 protein included in its effector domain prevents the covalent connection between ISG15 and its target proteins both in vitro and in infected cells.
