The Sulfate Assay Kit (SKU: BA0057) provides a simple, direct and automation-ready method for the quantitative turbidimetric determination of inorganic sulfate in biological fluids such as serum and urine. Inorganic sulfate is one of the most abundant anions in mammalian plasma and plays important physiological roles in activating and detoxifying xenobiotics, steroids, neurotransmitters and bile acids, as well as in the biosynthesis of glycosaminoglycans, cerebroside sulfate and heparin sulfate. In mammals, sulfate homeostasis is regulated by the kidney. The improved method utilises the quantitative formation of insoluble barium sulfate in polyethylene glycol, with the turbidity measured between 540 and 610nm being proportional to the sulfate level in the sample. The procedure involves addition of a single working reagent and incubation for 5 minutes, making it well suited to research and drug discovery applications.

This kit measures sulfate concentration in biological fluids such as serum and urine. The improved method utilises the quantitative formation of insoluble barium sulfate in polyethylene glycol, and the turbidity measured between 540 and 610nm is proportional to the sulfate level in the sample.

| Step | Procedure |
| 1 | Sample treatment: dilute urine 10-fold in deionised water prior to assay. Deproteinate serum or plasma by mixing 200 µL sample with 100 µL TCA Reagent, spinning down precipitates 5 minutes at 14,000 rpm and using 200 µL supernatant. |
| 2 | Standards: prepare a 2.0 mM Premix by mixing 20 µL 60 mM Sulfate Standard with 580 µL dH2O and dilute as shown in the table. Transfer 200 µL standards into wells of a clear flat-bottom 96-well plate (treat with TCA if deproteination is required). Transfer 200 µL samples into separate wells. |
| 3 | Prepare Working Reagent fresh (95 mg Reagent B per mL Reagent A), vortex at least 1 minute and incubate 10 minutes before use. Add 100 µL Working Reagent to each well and tap plate to mix. |
| 4 | Incubate 5 minutes at room temperature and read optical density at 540-610nm (600nm). |
Sulfate concentration = [(ODSAMPLE − ODH2O) / Slope] × n (mM), where ODSAMPLE and ODH2O are the OD values of the sample well and the water well (Standard #4), and n = 10 for urine. Conversion: 1 mM sulfate equals 9.61 mg/dL or 96.1 ppm.
