Ferroptosis is an iron-dependent, regulated form of cell death driven by the unchecked peroxidation of polyunsaturated phospholipids in cellular membranes, biochemically and morphologically distinct from apoptosis and necroptosis. The central defense against ferroptosis is the selenoenzyme GPX4, a glutathione-dependent peroxidase that reduces lipid hydroperoxides to nontoxic alcohols; its inactivation or loss permits lethal accumulation of oxidized lipids. GPX4 depends on a supply of reduced glutathione, the synthesis of which requires cystine import through the cystine-glutamate antiporter SLC7A11/xCT, making this transporter a key node controlling ferroptosis sensitivity. The execution of ferroptosis also depends on the lipid composition of membranes: ACSL4 preferentially incorporates polyunsaturated fatty acids into phospholipids, generating the substrates that are most prone to peroxidation.
