The Formaldehyde Assay Kit (Fluorometric) (SKU: BA0022) provides a convenient fluorimetric method for quantifying formaldehyde in biological samples. Formaldehyde is the simplest aldehyde and is naturally present in all tissues and body fluids, while also being widely used in industry and as a tissue fixative. Some cancers exhibit elevated formaldehyde levels, and increased urinary formaldehyde has been associated with prostate and bladder cancer, making its measurement a useful research tool. In this assay formaldehyde is derivatised with acetoacetanilide in the presence of ammonia, and the resulting fluorescent product is quantified at λex/em = 370/470 nm. The method is simple, sensitive and stable, detecting as little as 1.5 µM formaldehyde. Its homogeneous mix-incubate-measure format makes it well suited to high-throughput screening.

A homogeneous fluorometric assay for the quantitative determination of formaldehyde in biological samples. Formaldehyde is derivatised with acetoacetanilide in the presence of ammonia to yield a fluorescent product measured at λex/em = 370/470 nm. The assay detects as little as 1.5 µM formaldehyde.

| Step | Procedure |
| 1 | Standards: mix 5 µL of the provided 10 mM Formaldehyde with 495 µL dH2O to make a 100 µM Premix, then dilute as shown in the standards table. Transfer 50 µL standards into separate wells of a black flat-bottom plate. |
| 2 | Sample preparation: dilute urine 2-5 fold with dH2O and clear any particulates by filtration or centrifugation. Deproteinate high-protein samples with 50 µL 10% TCA per 100 µL sample, then neutralise 100 µL supernatant with 25 µL Neutralizer (multiply results by 1.875). |
| 3 | Add 50 µL of each prepared sample to two separate wells (one serves as a Sample Blank). |
| 4 | Prepare Working Reagent per well by mixing 33 µL Reagent A and 22 µL Reagent B (for Sample Blanks use 33 µL Reagent A + 22 µL dH2O). Add 50 µL to each well, tap to mix and incubate at room temperature for 30 min protected from light. |
| 5 | Read fluorescence intensity at λex = 370 nm and λem = 470 nm. |
Plot the RFU at 30 min for each standard against concentration and determine the slope by linear regression. [Formaldehyde] = (RFU_sample - RFU_blank - RFU_water) / slope x n (µM), where n is the dilution factor (1.875 for deproteinated samples). Conversion: 1 µM formaldehyde is equivalent to 30 ppb.
