The Liquid Zinc Assay (SKU: BA0051) provides a simple, direct and automation-ready method for measuring zinc directly in biological samples without any pretreatment. Zinc is an essential trace element required for the activity of more than 300 enzymes, the structure of many proteins and the control of genetic expression, and its status affects cell division, growth, differentiation and development. Zinc deficiency is commonly caused by low dietary intake and low bioavailability and is associated with conditions including malabsorption syndrome, chronic liver and renal disease, sickle cell disease, diabetes and malignancy. The method utilises a chromogen that forms a coloured complex specifically with zinc, with the intensity of the colour, measured at 425nm, directly proportional to the zinc concentration. Only 50 µL of sample is required, and the optimised formulation greatly enhances reagent and signal stability in cuvette or 96-well plate formats.

This kit measures zinc directly in biological samples without pretreatment. The method utilises a chromogen that forms a coloured complex specifically with zinc, with the intensity of the colour, measured at 425nm, directly proportional to the zinc concentration in the sample.

| 1 | Sample preparation: ensure serum and plasma samples are clear and free of turbidity; remove precipitates by filtration or centrifugation. Dilute serum or plasma 5-fold (n = 5) in deionised water before assay. |
| 2 | Equilibrate all reagents to room temperature and vortex Reagents B and C. Prepare Working Reagent: for each assay well mix 200 µL Reagent A, 4 µL Reagent B and 4 µL Reagent C. |
| 3 | Prepare standards in deionised water and transfer 50 µL of the Zn2+ standards into wells of a clear flat-bottom 96-well plate. Transfer 50 µL Sample and Sample Blank (50 µL sample + 2 µL EDTA) into wells. Add 200 µL Working Reagent to all wells and tap lightly to mix. |
| 4 | Incubate 30 minutes at room temperature and read optical density at 425nm (range 420-426nm). |
Subtract blank OD (water, #5) from the standard OD values and plot ∆OD against Zn2+ standard concentrations. Calculate ∆OD for the Sample (= ODSAMPLE − ODSAMPLE BLANK) and determine the Sample Zn2+ concentration from the standard curve by non-linear regression using a single-site saturation binding function. If Zn2+ exceeds 10 µM, dilute in deionised water, repeat the assay and multiply by the dilution factor. Conversion: 1 µM zinc equals 6.5 µg/dL or 0.065 ppm (65 ppb).
