Human Fc gamma Rs are divided into three classes, designated Fc gamma RI (CD64), Fc gamma RII (CD32), and Fco gamma RIII (CD16), which produce a variety of forms. The activating-type receptor is either linked noncovalently to an accessory subunit with an immunoreceptor tyrosine-based activation motif (ITAM) in its cytoplasmic domain or contains a non-covalent link to one. Fc gamma RI is a high-affinity receptor that participates in early immune responses, whereas Fc gamma RII and RIII are low-affinity receptors that bind IgG as aggregates surrounding multivalent antigens during late immune responses. The human CD32 protein; also known as Low affinity immunoglobulin alpha Fc region receptor II-a (IgG Fc receptor II-a); FCGR2A Protein or Fc alpha RII A or FCGR2A Proteindisplayed on cells of both myeloid and lymphoid lineages, CD32a (Fc gamma RII A) is the ITAM-bearing adapter subunit; FcR gamma; CD32a (Fc gamma RII A) sends an activating signal after ligand binding. This results in the initiation of inflammatory responses including cytolysis, phagocytosis, degranulation; and cytokine production. The responses can be modulated by signals from the co-expressed inhibitory receptors such as Fc alpha RII B; and the strength of the signal is dependent on the ratio of expression of the activating and inhibitory receptors.
