Choosing a kit

Which kit do I need?

Start with the label that fits a free channel on your instrument, or biotin if you plan to detect with streptavidin. Each label is then offered in versions that differ in the filtration tube supplied for purification, for example 3 kDa or 10 kDa cut-off, so choose the one suited to the size of your protein. The cut-off is stated under Applications on each product page.

The dyes react with primary amines, so remove Tris, glycine, sodium azide and carrier proteins such as BSA from the protein before labeling. A BSA removal kit is available in assay reagents.

Workflow

How labeling works

1

Prepare the protein

Exchange into an amine-free buffer and remove carrier protein.

2

Label

Mix with the reactive dye or biotin and incubate.

3

Purify

Remove unreacted label with the filtration tube supplied.

FAQs

Labeling kit FAQs

What can I label?

Antibodies and other proteins that contain primary amine groups.

Why are there several kits with the same dye?

They differ in the filtration tube supplied for clean-up, to suit proteins of different sizes. Check the Applications line on each product page.

Will labeling affect my antibody?

Over-labeling can reduce binding. Follow the recommended protein-to-label ratio and test the conjugate against the unlabeled antibody.

Which buffer components interfere?

Anything containing primary amines, such as Tris or glycine, as well as sodium azide and carrier proteins.
