The Magnesium Assay Kit (Colorimetric) (SKU: BA0045) provides a simple, direct and automation-ready method for measuring magnesium directly in biological samples without any pretreatment. Magnesium is one of the most abundant and essential minerals in mammals, involved in more than 300 biochemical reactions and playing important roles in muscle and nerve function, heart rhythm, the immune system and bone formation. Magnesium deficiency may lead to nausea, fatigue, muscle contractions, hypocalcaemia and hypokalaemia. The kit uses a calmagite dye that forms a coloured complex specifically with magnesium, and the intensity of the colour, measured at 500nm, is directly proportional to the magnesium concentration. As little as 5 µL of sample is required. The optimised formulation minimises interference by potential substances and can be run in cuvette or 96-well plate format.

This kit measures magnesium directly in biological samples without pretreatment. A calmagite dye forms a coloured complex specifically with magnesium, and the intensity of the colour, measured at 500nm, is directly proportional to the magnesium concentration in the sample.

| 1 | Prepare Working Reagent by combining equal volumes of Reagent A and Reagent B; equilibrate to room temperature before use. |
| 2 | Dilute Standard to 2 mg/dL by mixing 40 µL 10 mg/dL Standard with 160 µL distilled water. Transfer 5 µL diluted standard and samples in duplicate to wells of a clear bottom plate. |
| 3 | Add 200 µL Working Reagent and tap plate to mix thoroughly. |
| 4 | Incubate 2 minutes at room temperature and read OD at 500nm (sample and standard OD). |
| 5 | Add 10 µL EDTA Solution to all wells, tap to mix, incubate 2 minutes and read OD at 500nm (blank OD). |
Magnesium concentration = [(ODSAMPLE − ODBLANK) / (ODMG − ODMGBLANK)] × 2 (mg/dL), where ODSAMPLE and ODBLANK are OD500nm values of the sample before and after EDTA addition, and ODMG and ODMGBLANK are OD500nm values of the 2 mg/dL standard before and after EDTA addition. Conversion: 1 mg/dL Mg2+ equals 411 µM, 0.001% or 10 ppm.
