Mitochondria are dynamic organelles that continuously fuse and divide, and their morphology, number, and quality are matched to cellular energy demand through coordinated programs of fission, fusion, and biogenesis. Fission is executed by the cytosolic GTPase DRP1/DNM1L, which is recruited to the outer membrane and assembles into constricting spirals that sever the organelle, enabling mitochondrial distribution during division and the segregation of damaged segments for mitophagy. Opposing fission, fusion of the outer membrane is mediated by the membrane-anchored GTPases MFN1 and MFN2, which tether and merge adjacent mitochondria, while inner membrane fusion and cristae organization depend on OPA1; the balance between these forces shapes a network that ranges from fragmented to highly interconnected. Mitochondrial biogenesis is governed by the transcriptional coactivator PGC-1alpha, the master regulator that drives expression of nuclear-encoded respiratory genes and activates downstream factors controlling mitochondrial transcription. Among these, TFAM binds and packages mitochondrial DNA into nucleoids and is essential for its replication and transcription, directly linking biogenesis to the maintenance of the mitochondrial genome. The outer membrane proteins VDAC1, the principal channel for metabolite and ion exchange and a hub for apoptotic signaling, and TOM20, a receptor of the translocase of the outer membrane that imports nuclear-encoded precursors, serve as widely used markers of mitochondrial mass and integrity.
