alpha-2-macroglobulin, also known as alpha;2-macroglobulin (alpha;2M and A2M), is an abundant protein of the plasma of vertebrates and members of several invertebrate phyla and functions as a broad-spectrum protease-binding protein. The major protein component of the alpha-2 band in protein electrophoresis is alpha-2-macroglobulin, which is generated by the liver. A major plasma glycoprotein, alpha-2-macroglobulin has long been recognized as an irreversible proteinase inhibitor. It has been confirmed that a variety of growth factors, cytokines, and hormones bind to alpha 2M in various ways. A2M is also produced in the brain where it binds multiple extracellular ligands and is internalized by neurons and astrocytes. A2M has been found in the brains of Alzheimer's disease (AD) patients, where it is distributed throughout amyloid plaques. A2M binds soluble beta-amyloid and mediates its degradation. Because alpha2-macroglobulin and its protease cargo are selectively bound by cells in contact with the body fluids, they are then internalized and destroyed in secondary lysosomes of those cells. Alpha2-macroglobulin binds a variety of ligands, including several peptide growth factors and modulates the activity of a lectin-dependent cytolytic pathway in arthropods.
