Background

A sandwich immunoassay on a membrane

Each array is a membrane pre-spotted with capture antibodies, one position per target. The sample is incubated on the membrane and each protein is trapped by its matched capture antibody. Bound proteins are detected with a cocktail of biotinylated antibodies, followed by streptavidin-HRP and a chemiluminescent substrate. Spot intensity is proportional to the amount of protein and is quantified by densitometry.

The read-out is semi-quantitative: it shows which proteins change between samples and by how much, relative to a control. That makes an array a fast way to reach a shortlist of candidates from a whole pathway or disease signature, which can then be confirmed and measured in absolute terms by ELISA.

FAQs

Antibody array FAQs

What is a membrane antibody array?

A membrane carrying a grid of capture antibodies, each specific for a different protein. One incubation with the sample reports on every target on the membrane at once.

How many proteins can I detect on one array?

Between 7 and 274, depending on the array. The number of targets is given in each product name.

Is the data quantitative?

It is semi-quantitative. Arrays show relative changes in protein levels between samples. Use an ELISA to measure absolute concentrations of the hits.

Which sample types are compatible?

Serum, plasma, cell culture supernatants, cell lysates and tissue lysates. Check the product page for the samples validated for each array.

What equipment do I need?

A standard western blotting set-up and a chemiluminescent imaging system. No dedicated array reader is required.

Which size should I order?

Kits contain 2, 4 or 8 membranes. Each sample needs its own membrane, so count your samples and controls and include a replicate where you can.

How do I validate array hits?

Confirm candidates with a quantitative ELISA for each protein, or follow a set of them with a multiplex panel.
