Cell Culture Supernatant
Collect the culture supernatant.
Centrifuge at 1500 g for 10 minutes.
Concentrate the supernatant to approximately 0.5 mL using an ultrafiltration tube.
Filter the concentrated sample through a 0.45 µm filter before isolation.
Serum, Plasma, Breast Milk, Saliva
Centrifuge 1 mL of sample at 10,000 g for 10 minutes.
Carefully collect the supernatant.
Filter the supernatant through a 0.45 µm filter.
Use up to 0.75 mL of the filtered sample for isolation.
Urine
Collect 10–50 mL of first-morning midstream urine.
Centrifuge at 1500 g for 10 minutes.
Concentrate the supernatant to approximately 0.5 mL.
Filter through a 0.45 µm filter before isolation.
Plant
Extract approximately 35 mL of plant juice.
Incubate at 4 °C for 1 hour.
Centrifuge at 10,000 g for 10 minutes.
Concentrate the supernatant to approximately 0.5 mL.
Filter through a 0.45 µm filter before extraction.

Protocol

Choose the protocol based on the exosome sample type.
Serum or plasma samples: Transfer 500 µL of processed sample to a 1.5 mL centrifuge tube, add 500 µL Solution A, mix well, then add 250 µL Exosome Precipitation Solution and invert to mix thoroughly. If adjusting the sample volume, maintain the ratio Sample : Solution A : Precipitation Solution = 2 : 2 : 1.
Cell culture supernatant or urine samples: Transfer 1 mL of processed sample to a 1.5 mL centrifuge tube, add 250 µL Exosome Precipitation Solution and mix by inversion. If the sample volume is large, concentrate it to 1–2 mL before starting the procedure. When adjusting volumes, maintain the ratio Sample : Exosome Precipitation Solution = 4 : 1.

Incubate at room temperature for 30 minutes (rapid protocol). If time permits, incubate overnight at 4°C for higher recovery.

Centrifuge at 4°C, 12,000 × g for 30 minutes and discard the supernatant. After discarding or carefully aspirating the supernatant, use a 200 µL pipette to remove any remaining liquid.

Add 200 µL of Solution A to the exosome pellet and gently pipette up and down to fully resuspend the precipitate.

Transfer the suspension to a new 1.5 mL centrifuge tube and centrifuge at 4°C, 12,000 × g for 5 minutes. Retain the supernatant containing the exosome particles. If a large pellet remains, repeat centrifugation at 12,000 × g for 5 minutes until no significant pellet is visible.

The isolated exosomes can be used immediately for downstream experiments. If not used within 24 hours, store at 4°C. For longer storage, aliquot and store at −80°C.
