Granulocyte Colony Stimulating Factor Receptor (G-CSFR), also known as CD114, is a cell surface receptor for colony stimulating factor 3 (CSF3). G-CSFR is a key regulator of granulopoiesis. The extracellular region of this type I membrane protein is made up of an immunoglobulin-like domain, a cytokine receptor-homologous (CRH) domain, and three fibronectin type III domains. G-CSF receptor mutations causing carboxy-terminal truncation transduce hyperproliferative growth responses and are associated with the invasive and malignant progression of severe congenital neutropenia (SCN) to acute myelogenous leukemia (AML). G-CSFR activation may also be important in the biology of solid tumors, particularly metastasis, through autocrine/paracrine stimulation.
