Spike & Recovery

In order to asses the accuracy of an ELISA kit, spike and recovery and linear of dilution experiments provide important information in the validation and assessment of the accuracy of the ELISA kit assay.

The use of spike and recovery allows researchers to determine whether the detection of their analyte of interest is affected by the standard diluent and the biological sample matrix. In a spike and recovery experiment to assess and validate and ELISA kit a known amount of analyte (recombinant protein or hormone) is added into a test sample matrix and the response is compared to the identical spike into the standard diluent.

The example below of matrices listed below were spiked with certain level of IL8 and the recovery rates were calculated by comparing the measured value to the expected amount of IL8 in samples.

In order to assess the precision for samples tested at different dilutions a linearity of dilution experiment is carried out. If the linearity of the assay is good over a range of dilutions (i.e. 1:2 to 1:16 for example), then the assay provides the ability to assay a sample with different levels of analyte. If a sample contains a high level of analyte, a sample may be diluted with standard dilution buffer and allow for comparison with an undiluted sample that contains low levels of the analyte of interest.

In the example below the linearity of the kit was assayed by testing samples spiked with appropriate concentration of IL8 and their serial dilutions. The results were demonstrated by the percentage of calculated concentration to the expected.

Coefficient of Variability (% CV)

ELISA kits are great tools to allow researchers to quantifiably measure the amount of analyte in a sample. However, in order to assess and show how well and ELISA assay kit is ran the results of the assay must a low coefficient of variability (%CV or CV). Whereby the %CV statistically shows the variance between replicate samples (Intra-assay) in an assay or between separately run assays (Inter-assay).
