While conjugates of this protein have been found to be noncovalently attached to intermediate filaments, this protein is sometimes secreted. Western blot analysis of lysates from Jurkat cells, using ISG15 Rabbit pAb (CAB1182) at 1:1000 dilution. Jurkat cells were treated with IFN-α (100 ng/ml) at 37℃ for 30 minutes after serum-starvation overnight. Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) (AS014) at 1:10000 dilution. Lysates/proteins: 25μg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit (AbGn00020). Exposure time: 5s.
