Cellular senescence is a stable proliferative arrest enforced by the p16^INK4A-Rb and p53-p21 axes, accompanied by loss of Lamin B1 and a senescence-associated secretory phenotype (e.g. IL-6, HMGB1). Senescence is central to ageing, tumour suppression and therapy response.
